    摘要:

    目的:探讨瑞香素诱导人胶质母细胞瘤U-251细胞自噬的机制。方法:用不同浓度瑞香素处理U-251细胞48 h后,采用透射电镜观察U-251细胞内自噬体的数量及形成情况;Western blot法检测并比较自噬相关蛋白mTOR、P62、Beclin-1、LC-3的表达水平。结果:与对照组比较,40、80和160μg/mL的瑞香素处理48 h后,U-251细胞后内自噬体的数量均增加(P<0.05);mTOR、Beclin-1和LC-3蛋白的表达水平均升高(P<0.05);P62蛋白的表达水平均降低(P<0.05),且呈剂量依赖性。结论:瑞香素可能通过上调mTOR、Beclin-1和LC-3蛋白的表达、下调P62蛋白的表达以促进U251细胞自噬发生。

    Abstract:

    Objective:To investigate the effect and mechanism of daphnetin-induced autophagy in human glioblastoma U-251 cells.Methods:After U-251 cells were treated with different concentrations of daphnetin for 48 h,the number and formation of autopha-gosomes in U-251 cells were observed by transmission electron microscopy.Western blot was used to detect the expression of autophagy-related proteins mTOR,P62,Beclin-1 and LC-3.Results:Compared with the control group,the number of autophagosomes in U-251 cells was increased after treatment with 40,80 and 160 μg/mL daphnetin for 48 h.The expression levels of mTOR,Beclin-1 and LC-3 proteins were increased,while the expression level of P62 protein was decreased in a dose-dependent manner.Conclusion:Daphnetin may promote autophagy in U251 cells by up-regulating the expression of mTOR,Beclin-1 and LC-3 proteins and down-regulating the ex-pression of P62 protein.
