    摘要:

    目的:探讨冬凌草甲素介导NLRP3炎症小体对戊四氮慢性癫痫模型小鼠发作易感性的调控作用。方法:利用不同剂量Ori治疗戊四氮诱导的慢性癫痫小鼠,观察治疗前后小鼠行为学(癫痫发作等级、发作潜伏期及频率),并用HE染色检测治疗前后小鼠海马组织细胞形态,Western blot检测NLRP3及相关蛋白表达水平。结果:行为学结果显示,冬凌草甲素治疗后能延长癫痫发作潜伏期,改善癫痫发作严重程度,降低癫痫自发发作频率,特别是中高剂量治疗组。HE染色、Western blot检测结果与行为学结果一致,经Ori治疗后能一定程度减轻癫痫引起的病理损伤,减少炎症浸润,降低颅内NLRP3及相关蛋白表达水平。结论:冬凌草甲素调控癫痫易感性,可能是通过抑制NLRP3炎性小体通路介导,说明Ori对癫痫有一定的神经保护作用。

    Abstract:

    Objective:To explore regulatory effect of oridonin(Ori)-mediated NLRP3 inflammasome on seizure susceptibility in PTZ-induced chronic epileptic mice.Methods:Chronic epilepsy was induced in mice using pentylenetetrazol(PTZ),and the mice were treated with different doses of Ori.Behavioral changes(seizure severity,latency,and frequency)were observed before and after treat-ment.Hippocampal tissues were examined using HE staining,and the expression levels of NLRP3 and related proteins were detected by Western blot.Results:Behavioral results showed that Ori treatment prolonged seizure latency,reduced seizure severity,and decreased the frequency of spontaneous seizures,particularly in the medium-and high-dose groups.HE staining and Western blot results were con-sistent with the behavioral findings.Ori treatment alleviated epilepsy-induced pathological damage,reduced inflammatory infiltration,and downregulated the expression levels of NLRP3 and related proteins in the brain.Conclusion:Ori may regulate epilepsy susceptibility by inhibiting the NLRP3 inflammasome pathway,suggesting that Ori has a protective effect against epilepsy.
