    目的:探讨头穴丛刺对淀粉样前体蛋白/早老素1双转基因(APP/PS1)小鼠神经炎症及酪氨酸激酶2/信号转导 及转录激活蛋白3信号通路(JAK2/STAT3)的影响。方法:将18只雄性 APP/PS1转基因小鼠随机分为模型组与头穴丛刺 组,每组各9只;并选取9只同月龄 C57BL/6小鼠作为空白组。头穴丛刺组每日在“百会穴”及其左右2mm 旁开处进行针刺 1次,持续28d。使用莫里斯(Morris)水迷宫评估小鼠的学习与记忆能力;尼氏染色法用于观察海马神经元数量;酶联免疫吸 附测定(ELISA)检测血清白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)水平;Westernblot分析海马中 JAK2、p-JAK2、STAT3及 p-STAT3蛋白的表达。结果:与空白组相比,模型组小鼠的逃避潜伏期延长,穿越平台次数和目 标象限停留时间减少(P<0.01);海马神经元数量降低(P<0.01);血清IL-6、IL-1β、TNF-α水平均升高(P<0.01);p-JAK2 及p-STAT3蛋白的表达水平上升(P<0.01)。与模型组相比,头穴丛刺组小鼠认知功能改善(P<0.05);海马神经元数量增 多(P<0.01);血清IL-6、IL-1β、TNF-α水平均降低(P<0.01);p-JAK2及 p-STAT3蛋白表达下降(P<0.01)。结论:头穴 丛刺可能通过调节JAK2/STAT3信号通路抑制神经炎症反应,从而改善 APP/PS1小鼠的认知功能。

    Abstract:

    Objective: To explore the effects of scalp cluster acupuncture on neuroinflammation and the Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) signaling pathway in amyloid precursor protein/presenilin 1 double transgenic (APP/PS1) mice. Methods: 18 male APP/PS1 transgenic mice were randomly divided into model group and scalp cluster acupuncture group, with 9 mice in each group, and 9 age-matched C57BL/6 mice were selected as the blank control group. Mice in the scalp cluster acupuncture group received acupuncture once daily at Baihui (GV20) and two points 2 mm lateral to the left and right of Baihui for 28 consecutive days. Morris water maze was used to evaluate learning and memory ability of mice. Nissl staining was used to observe the number of hippocampal neurons. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum levels of interleukin-6 (IL-6), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α). Western blot was used to analyze the expression of JAK2, p-JAK2, STAT3 and p-STAT3 proteins in the hippocampus. Results: Compared with the blank control group, the escape latency of mice in the model group was prolonged, and the number of platform crossings and the time spent in the target quadrant were reduced (P<0.01), the number of hippocampal neurons was decreased (P<0.01), serum levels of IL-6, IL-1β and TNF-α were increased (P<0.01), and the protein expression levels of p-JAK2 and p-STAT3 were increased (P<0.01). Compared with the model group, cognitive function of mice in the scalp cluster acupuncture group was improved (P<0.05), the number of hippocampal neurons was increased (P<0.01), serum levels of IL-6, IL-1β and TNF-α were decreased (P<0.01), and the protein expression levels of p-JAK2 and p-STAT3 were decreased (P<0.01). Conclusion: Scalp cluster acupuncture may inhibit neuroinflammatory responses by regulating the JAK2/STAT3 signaling pathway, thereby improving cognitive function in APP/PS1 mice.
