    Abstract:

Background : Hypopharyngeal squamous cell carcinoma is one of the malignant tumors with high incidence in the head and neck. It has the characteristics of strong invasiveness, early occult metastasis, and clinical diagnosis mostly in the middle and late stages. At present, the comprehensive treatment plan based on surgery combined with radiotherapy and chemotherapy has limited benefits. It is urgent to explore the key molecules that regulate tumor progression as potential targeted therapeutic targets. Objective: To explore the influence of CHMP4C on the proliferation, migration and invasion abilities of Fadu cells in hypopharyngeal squamous cell carcinoma. Methods: Fadu cell model with stable knockdown of CHMP4C (shCHMP4C group) and negative control group (shNC group) were constructed. Western Blot was applied to verify the knockdown efficiency of CHMP4C protein. CCK-8 assay was utilized to detect the cell proliferation activity. Transwell migration assay and Matrigel invasion assay were adopted to evaluate the cell migration and invasion abilities. The expression level of epithelial-mesenchymal transition (EMT) key marker E-cadherin was detected by Western Blot. Results: Western Blot analysis revealed significantly reduced CHMP4C protein expression in the shCHMP4C group compared to the shNC group (P<0.05). CCK-8 assay demonstrated markedly slowed cell proliferation in the shCHMP4C group, with lower OD values at 48 h and 72 h compared to the shNC group (P<0.05). Transwell migration and invasion assays suggested reduced transmembrane cell counts in the shCHMP4C group compared with those in the shNC group (P<0.05). Western Blot detection found upregulation of E-cadherin protein expression in the shCHMP4C group relative to the shNC group (P<0.05). Conclusion: CHMP4C can promote the proliferation, migration and invasion abilities of Fadu cells in hypopharyngeal squamous cell carcinoma, which may be achieved by regulating the EMT process. CHMP4C may serve as a potential candidate molecular target for targeted therapy of hypopharyngeal squamous cell carcinoma, although its clinical value requires further investigation.
